Mechanisms of Gene Regulation

Identification of QTL which will regulate the proportions of saturated and unsaturated fatty acids in seed oils of Arabidopsis thaliana.

Quantitative trait loci (QTL) that control fatty acid composition in seed oils in Arabidopsis thaliana were studied using recombinant inbred population derived from a cross between the Arabidopsis ecotypes Landsberg erecta and Shakdara. Composite QTL model mapping identified 12 major QTL and 12 minor QTL. The most significant QTLs are located on chromosomes 3 and 4. There are strong QTL for oleic acid (18:1) and linoleic acid (18:2) which colocate with the Stearoyl-ACP desaturase and FAD2 loci located on the upper arm of chromosome 3. There are other strong QTLs for eicosenoic acid (20:1), eicosadienoic acid (20:2) and erucic acid (22:1) that colocate with Fatty acid elongase1(FAE1) locus on lower arm of chromosome 4. Fine mapping will be done to identify possible genes where major QTLs have been identified which do not colocate with candidate genes. Further studies will be done to identify QTL which account for genetic variation in fatty acid composition in populations.
