Secondary Metabolism

The study of coumarins production after elicitation in Angelica gigas suspension culture.

Industrial application of plant cell system is limited by low yields of target compounds and high cost of cell culture system, for example long culture and bioreactor. Many researchers have tried to overcome these limitations by optimization of plant cell systems and molecular biotechnology. But characterization of many medicinal plants was not still revealed. To enhance secondary metabolite productivity, characterization of medicinal plant must be investigated above all. Analysis of secondary metabolite and gene expression level related with biosynthetic pathway are very important.
Angelica gigas Nakai produces a variety of coumarins of pharmaceutical importance, including the decursin, decursinol angelate, nodakenin and ¥â-sitosterol. Angelica gigias has been used as a traditional Korean herbal medicine which has novel cancer chemotherapeutic compounds. Biosynthetic pathways of coumarin derivatives are associated with phenylalanine metabolism containing phenylalanine ammonia-lyase.
Elicitor treatment is a good tool for increasing productivity of secondary metabolites and revealing unexpected new secondary metabolites. We activated all secondary metabolisms by the addition of biotic elicitors to suspended cultured cells and compared secondary metabolites between control (unelicited) cells and elicited cells. Elicitor-treated cells showed higher productivity of coumarin and coumarin derivatives than untreated cells. Also Elicitor-treated cells produced secondary metabolites which are not shown in unelicited cell by opening other secondary metabolism. We investigated major relationship between profiles of metabolite production and expression level of phenylalanine ammonia-lyase (PAL). It gave us important information on characterization of traditional Korean medicinal plant in coumarin production pathway.
