Contribution of pollen ontogeny to the phylogeny of Papaveraceae s.l.: Congruence between ontogenic and molecular data.

Morphological characteristics of mature pollen grains have been traditionally used as a source of characters for plant taxonomic studies. In Papaveraceae, previous studies have shown that many different pollen types exist, and that the morphological characters observed in mature grains appear homoplasious. In contrast, the utility of pollen wall ontogenic ultrastructural characters has not been assessed in this group, and, indeed, such characters have rarely been used in phylogenetic reconstruction. Here we assess variation in ontogenic characters, and compare phylogenies derived from these characters and from a molecular phylogeny of Papaveraceae and related families using the chloroplast region trnL-F. The molecular phylogeny reflects the main groupings found within Papaveraceae s.l. on the basis of earlier morphological and molecular analyses, and, although data from more taxa are needed, the phylogeny based on ontogenic analysis was highly congruent with that obtained from the molecular analysis. This confirms the phylogenetic utility of the pollen-wall ontogenic characters and suggests that analysis of pollen wall ontogeny is a powerful source of data to clarify relationships amongst early eudicots. Furthermore, molecular and ontogenic analyses helped us establish the evolutionary direction of important characters in the pollen ontogeny of this early eudicot family, as for example the columellar infratectum (ancestral in Papaveraceae s.l.) and the lamellate foot-layer of the ectexine (a derived character).
