Changes in macromolecule composition and the levels of metal ions in the early stages of salt stress in two cultivars of sweet potato under in vitro conditions. Salinity stress is one of the major abiotic stresses affecting sweet potato cultivation. Our preliminary results showed that there was fluctuation in the plant growth between early and later stages of exposure to salt stress. The present work was carried out to determine the immediate effects of salt stress on two cultivars of sweet potato [ Ipomoea batatas L.] Salyboro and Commensal. These were grown under in vitro conditions for 6 weeks to allow them to reach a size of six nodes. At this stage, the plantlets were removed and reintroduced to media containing different NaCl levels ranging from 0 to 200mM. One set of plantlets were removed at 24 hrs and the other set at 48 hrs. Roots, leaves and stems were analyzed to determine the uptake of mineral elements [ Na, K, Mg, Ca, Fe, Cu, Zn, Co, Mn] and to changes in the levels of soluble carbohydrates, proline and protein levels. In both cultivars, the uptake of sodium was high in the first 24 hours, increasing the concentration in the root and stem by almost ten-fold . Both potassium and magnesium levels did not change in the root but increased in the stem while the leaves showed decreased levels. The levels of calcium were severely reduced in both the root and the leaf but showed slight increase in the stem. The most significant effect was observed in Fe levels with very severe reduction in the root, stem and leaf. Zinc concentrations did not change much. The other elements had mixed trends. Soluble carbohydrates and proline showed increase in the stem and leaf but not in the root while protein concentrations decreased slightly. By 48 hrs, there was some recovery, especially with calcium ions but Fe was still low. These results suggest that Fe uptake may have an important role in adjusting to salt stress.
