Development of a method for localization of low concentrated heavy metals in plant tissues.

For the understanding of the biological and ecological effects of heavy metals the precise localization of them in plant tissues is essential. Most techniques used hitherto are however successfully applicable only for hyperaccumulators plants. For other plant species the tissue concentration is below the detection limit of most equipments. Therefore the metal distribution and time resolved insights in the transport phenomena are still badly understood in both types of non-hyperaccumulators, the indicators as well the excluders plants. In the present work, we suggest the use of the laser ablation coupled to ICP-MS (LA-ICP-MS) as a promising technique to localize heavy metal in plant tissues with low concentration of metal. Nickel localization in the leaf tissues of Ricinus communis was investigated. The plants were cultivated in nutrient solution with nickel addition (60 µM) and, after 40 days of exposition, leaves were collected, air dried, and analysed by LA-ICP-MS. The spatial resolution is still a relevant limiting factor; however the laser ablation technique combined with anatomical analysis after ablation has permitted to verify differences in nickel distribution in parenchyma. Therefore this technique may lead to a much better understanding of the distribution the metal in plant non-hyperaccumulators.
