Population Genetics Huang, Daisie [1], McDade, Lucinda A. [2], Friar, Elizabeth [3]. Chloroplast inheritance in Lupinus. We are investigating the chloroplast inheritance patterns of two closely related lupines, Lupinus nanus and Lupinus bicolor. While strict maternal inheritance of chloroplasts is generally assumed to be true for the majority of angiosperms, heteroplasmy and biparental inheritance of chloroplasts have been documented in members of the family Fabaceae (Leguminosae) as well as in other genera and families. While heteroplasmy at an intraspecific level is probably not significant enough to affect phylogenetic studies at the species level, heteroplasmy at the population level may confound genetic and phylogenetic studies of taxa that do not have clear species boundaries and may be sharing genes, even at low frequencies. To investigate the impact possible paternal or biparental plasmid inheritance may play on chloroplast phylogenetics in the genus Lupinus, we are conducting a population-level screen of populations of sympatric L. nanus and L. bicolor. We are using PCR amplification of noncoding chloroplast regions, followed by restriction enzyme digestion and fragment analysis, to gather data about the plastid haplotypes in the study populations and examine any population-level variation.
