Inheritance bias of the internal transcribed spacer of the nuclear ribosomal DNA in artificial hybridization of Acer albopurpurascensxA. buergerianum var. formosanum. In the field, the two endemic species, A. albopurpurascens and A. buergerianum var. formosanum grow in the same habitat of north Taiwan. In their morphological characters, the leave variation of A. albopurpurascens is oblong-lanceolate to shallowly 3-lobed, which is similar to A. buergerianum var. formosanum. To understand the inheritance of ribosomal DNA and chloroplast DNA in artificial hybridization, experiments were conducted using A. albopurpurascens x A. buergerianum var. formosanum as paternal and maternal donors, respectively. The internal transcribed spacer region (ITS) and 5.8s of nuclear ribosomal DNA and the rbcL-atpB spacer in chloroplast non-coding region were amplified from the artificial hybrids and parental species. In the cpDNA data, the artificial hybrids were the same inheritance to maternal donors, A. buergerianum var. formosanum. In contrast to the single type ITS and 5.8s rDNA in the parental species, multiple sequences were cloned from artificial hybrids and their parental species. Sequencing lengths ranged from 627 to 648 bps in the ITS and 5.8s of nuclear ribosomal DNA. Nucleotide diversity was detected with the values of 0.00468 (pi) in A. buergerianum var. formosanum, 0.00959 in A. albopurpurascens, and 0.01956 in artificial hybridization, showing higher levels of genetic variation in the artificial hybrids. A split decomposition network based on ITS nucleotide and 5.8s variations revealed that all but three (clone F4-5, F5-2 and F6-1) of the hybrid sequences were "phylogenetically" closely related to parental species. The higher genetic recombination events (Rm =8) were observed in the artificial hybrids than A. albopurpurascens (Rm =2) and A. buergerianum var. formosanum (Rm =0). As the results above, the artificial hybrids showed the lineage sorting from the parental species, and a high level of genetic variation and recombination events were detected on the artificial hybrids. *All authors contributed equally to this work.
